Summary
Sphaerothecum destruens, also known as the rosette agent, is one of the most important intracellular parasites of fish. The genus Sphaerotecum has only two species, S. destruens and S. caipira, and is currently classified in clade Opisthokonta, class Ichthyosporea (Mesomycetozoea). Such taxonomy place it close to metazoa and fungi, but the term ‘protist’ or ‘fungus-like’ is used to refer to Ichthyosporeans, which includes Dermocystida order, comprising several other parasites of amphibians and fishes. S. destruens, for instance, causes diseases in salmoniformes (salmons) and cypriniformes (carps), a problem that is expected to cause serious economic damage. Nonetheless, genomic information for S. destruens is extremely scarce, while no genome or transcriptome assembly exist for this parasite. Therefore, in the present report the first de novo transcriptome assembly and functional annotation of Sphaerotecum destruens is brought to light. The de novo transcriptome of S. destruens was assembled with Trinity and characterized comprising of 12,000 genes and 19,720 transcripts, with 42.35 GC%. Contigs assembled have median and average length of 756 and 1,086, respectively, in a total of 21,415,945 assembled bases. Transcriptome completeness assessed by Benchmarking Universal Single-Copy Orthologs (BUSCO), identified 76.8% of the universal complete genes in Eukaryote database. With the aid of Eukaryotic Non-Model Transcriptome Annotation Pipeline (EnTAP), 13.624 (69%) elements received functional annotation from NCBI's nr and RefSeq databases and from EggNOG database: 13.066 (66%) annotated to protein domains; 12.493 (63%) were assigned at least one Gene Ontology; 11.942 (60%) to Biological Process; 10.316 (52%) to Cellular Component; and (58%) to Molecular Function.
