Summary
Microcystis cyanobacteria are able to naturally produce various types of microviridin and cause formation of toxic flowers in freshwater lakes. Members of this group have a potential serine protease inhibitor. An example is Microviridine B with high therapeutic potential as an elastase inhibitor in the treatment of pulmonary emphysema, which opens the door to biotechnological engineering. Due to the biotechnological potential, the goal of this study was to identify the agglomerates of biosynthetic microvirin genes in the cyanobacterial genome Microcystis aeruginosa using bioinformatic analysis. The M. aeruginosa strain CACIAM 03 whole genome was submitted to antiSMASH 4.0, in FASTA format using the "all on" mode of extras. An total of 42 aggregates were attributed. Among them, two clusters were related to biosynthesis of microviridina (clusters 25 and 42). The first one gene cluster showed 46% and 25% of similarity to the entire genome of M. aeruginosa TAIHU98 contig3 and with Planktothrix agardhii Microviridin K biosynthetic gene cluster, respectively. The second one demonstrated 60% and 53% similarity to the complete genome of M. aeruginosa SPC777 contig000278 and with M. aeruginosa NIES-843 DNA, respectively. Additionally, this gene cluster obtained 100% and 60% similarity with agglomerates of Microviridin B biosynthetic genes from two strains of M. aeruginosa and 80% similarity with the biosynthetic genes of Planktothrix agardhii Microviridin K. In the gene cluster 42, all genes involved in the biosynthesis of microvirin B were identified in the opposite sequence of the organism deposited in the database, being the MdnE sequence (100% gene similarity), mdnD (98%), mdnC (99%), , mdnB (98%) and mdnA (90%). These results demonstrate the M. aeruginosa strain CACIAM 03 biosynthetic potential in producing microviridine that can be applied in future biotechnological studies.
