Summary
Tityus serrulatus (scorpion) venom induces Reactive Oxygen Species production by RAW macrophages Introduction: Macrophages present an essential role against pathogens. Reactive oxygen species (ROS) produced by those cells controls several infections, since hydrogen peroxide, for example, has an important microbicidal and inflammatory function. In this work, we evaluated the effect of Tityus serrulatus venom (Tsv) on H2O2 production. Material and Methods: RAW 264.7 cells (1 x 106 / mL) were incubated with T. serrulatus venom (Tsv - 0.1, 1.0, 10.0 µg / mL) and / or LPS (10 µg / mL), for 24 or 48 hours. After treatments, cells were incubated with DHR (375 ng / mL) for 10 minutes to proceed H2O2 production assay on the flow cytometer. The experiments were performed in triplicate, and the one-way ANOVA test was applied, (p < 0.05). Results and Discussion: An increase in oxidative burst is an important feature of activated macrophages. The treatment with Tsv was able to induce H2O2 production, presenting significant values for all tested concentrations, both in 24 and 48 hours. However, Tsv / LPS associated treatment presented lower oxidative response, when compared to Tsv-stimulated groups, even this reduction was not statistically significant. Conclusion: T. serrulatus venom was able to activate macrophages by increasing ROS production. Thus, this work opens new perspectives for the use of T. serrulatus venom, which may lead to the development of new therapies for infections control.
