Summary
It has been shown that blue light (BL) perception by cryptochromes (crys) is an important upstream pathway of ABA or GA action. Thus, we hypothesized that cry1a of tomato modulates pivotal ABA and GA metabolism genes in the seed germination (SG). From previous results, we treated the seeds of tomato mutant cry1a and its control (Wild-type; WT), cv. Moneymaker, with continuous 10 µmol m-2s-1 BL or dark for 36 or 72 hours (h) in chamber growth, at 25 oC. Compared to dark at 36 h, seeds of WT and cry1a in BL condition showed enhanced relative expression (RE) of genes related to ABA synthesis, SlNCED1, whereas SlGA20-ox2, SlGA20-ox3 and SlGA3-ox2 (GA synthesis) were reduced. This seems to be dependent on the type of ABA and bioactive GA because in both light conditions SlNCED2 did not differ between genotypes, and SlGA20-ox1 was enhanced at 36 h in WT. Even at 36 h, the role of cry1a on hormonal control of SG became evident because compared to WT, the mutant reduced the RE of SlNCED2 and SlGA20-ox1 in the dark and BL, and SlGA20-ox2 and SlGA3-ox2 in the dark, whereas NCED1 and GA20-ox2 were enhanced in the dark and BL, respectively. At 72 h, RE of SlNCED1 of WT was higher than cry1a in BL, but in this condition, SlGA20-ox3 and SlGA3-ox2 were much enhanced compared to WT. Regarding GA inactivation genes, in BL SlGA2-ox4 and SlGA2-ox5 of cry1a were enhanced and reduced, respectively, compared to WT, indicating an intricate molecular mechanism of interaction between cry1a, ABA and GA over SG of tomato.
