Summary
The RNA-binding proteins (RBPs) are responsible for regulating an extensive number of genes by interacting with the RNA transcripts and regulating the fate and function of the bound RNAs. It is known that mutations in RBPs or changes in their regulation are associated with diverse disease types. The Bicaudal C homolog 1 (BICC1) is an RBP characterized by three K Homology domains in the N terminal end (KH1, KH2 and KH3 domains), which were related to the RNA-binding activity, and a Sterile Alpha Motif at the C-terminal end (SAM domain), which is related to the promotion of the protein-protein interaction. This protein has been mostly related by its role in polycystic kidney disease (PKD), kidney organogenesis and some kinds of cancer. We are currently characterizing the BICC1 protein interactors by Co-IP-MS and the BICC1-bound mRNA targets by CLIP-seq. We have compared the interactome in dependence of RNAs and in the presence and absence of the SAM domain. The BICC1 interacting proteins in the absence of RNA were mostly enriched in the ribosomal class while the ribosomal interactors were absent when RNA was present. We are following up the interaction with the ribosome. In this study, proteomics and bioinformatic analyses associated with molecular biology are guiding the interactome validations. Related the mRNA targets, CLIP-seq showed enrichment in 3’UTR and CDS of target mRNA. We have selected four mRNA targets related to the Wnt pathway and one microRNA and are currently testing the role of those interactions in cell culture. From this perspective, we are studying the role of BICC1 in cell signaling and microRNA regulation and we are guiding the work to further characterize BICC1 as drug target.
