Summary
Accidents associated with envenomation by Loxosceles spiders in South America should not be neglected. In Brazil, three species are most important in the context of public health: Loxosceles gaucho, L. intermedia and L. laeta. The composition of the venom of these spiders is complex, and the great diversity of components make to fully understand its mechanism of action difficult. Among these components, the Phospholipase D family of toxins plays a fundamental role in the clinical evolution of loxoscelism, and when isolated can reproduce the main effects of this pathology, such as dermonecrosis, exacerbated local inflammatory response and acute renal failure. The Phospholipases D compose a highly conserved family of enzymes in different venoms of Loxosceles and several isoforms may be present in the venom of distinct species. Due their importance, these toxins are a target for the study of loxoscelism. Although several isoforms of these toxins have already been described in the venoms of L. gaucho, L. intermedia and L. laeta, probably still exist many isoforms that have not been described due to variation in the expression profile of these animals according to different factors. Furthermore, the advancement of high throughput sequencing techniques, the identification of transcripts expressed at low levels has become more feasible. Therefore, to identify potential new phospholipases D expressed by Loxosceles venom gland, cDNA libraries were constructed for each species using next-generation sequencing (NGS) by MiSeq sequencer (Illumina). The processing of these samples and analysis of results become possible to identify transcripts coding for Phospholipases D not described until now. They are potential mature amino acid sequences similar to other Phospholipases D in Sicariidae family, but which have not been described for the sampled species. These data will be essential to identify structural and functional differences between proteins from the same family e will serve as a basis for future studies aimed at functionally evaluating these enzymes. The description of new isoforms for each species can help in the immunogenic and toxicological comparison between species of the same genus, contributing to the knowledge about the mechanism of action of Phospholipases D. The data obtained from these analyses will also be able to establish the evolutionary relationship between genes encoding Phospholipases D described in Sicariidae spider venoms at now.
