Resumo
Emerging pollutants have been targeted in recent researches for the potential risk they cause to human health and the environment. The management, application and irregular disposal of these products can lead to contamination of the environment, favoring their consumption through food and water. Among these pollutants are the pesticides, necessary in large-scale plantations, and the metal nanoparticles, which are applied in several areas, such as health and photonics. The pesticides used in this work were acephate, an insecticide of the organophosphorus family that can cause negative effects on the human neurological system, and diuron, a urea-class herbicide that has carcinogenic potential, especially in the bladder region. In the case of the metal nanoparticles, although they do not have regulation in their use, they can penetrate the cellular tissues, altering the properties of cell membranes. An important effect to consider is the mixture of various pollutants, which can lead to the "cocktail effect", resulting in different effects of these products in human health. To study these effects, one of the most used research directions is to investigate the biological action of these contaminants through the interaction of such substances with simplified models of the cell plasma membrane. This structure, in addition to be responsible for the cell integrity, is the way for substances to enter in the cells. In this work were investigated the effects, separately and together, of the pesticides acephate and diuron and gold nanoparticles in ternary giant unilamellar vesicles (GUVs) formed by the phospholipid POPC, sphingomyelin and cholesterol in the proportion found in the cell membrane (POPC/cholesterol/sphingomyelin – 70/20/10 mol%). First, the GUVs were formed by electroformation, hydrating the films of the lipid mixture deposited in platinum wires with sucrose solution in HEPES (0.2 M sucrose, 10 mM HEPES, pH adjusted to 7.4). After hydration, an electrical voltage of 1 V with a frequency of 10 Hz was applied in the system at temperature of 60 ºC for 1 hour. After the end of electroformation, the vesicles were analyzed by phase contrast microscopy for 1 hour in glucose solutions (0.2 M) in the absence and presence of acephate (1.28 x 10-4 M), diuron (1.28 x 10-4 M) or gold nanoparticle (4.28 µM), as well as the mixture of the three pollutants, all diluted in 10 mM HEPES solution (pH 7.4). During the analysis, images of the vesicles were obtained at 30 and 60 minutes, which were later counted considering the total of GUVs present in the images, as well as the total of these structures that presented morphological changes, such as non-spherical shape, presence of protrusions or loss of phase contrast. No significant morphological changes were observed through the method applied in the presence of pollutants, separately or together, indicating that, at the concentrations used, there are no effects of pesticides or nanoparticles, as well as their mixtures, on the vesicles.
