Summary
The investigation of new materials with multiple applications is a topic of great interest in technology and materials science. In this context, spectroelectrochemistry has emerged as a valuable tool for understanding mechanisms involved in electrochemical processes, as well as for the accurate determination of analytes, thus avoiding misinterpretation of electrochemical or spectroscopic data. This study reports on the development of a sensor platform for electrochemical and surface-enhanced Raman scattering (SERS) analysis, utilizing screen-printed carbon electrodes (SPCE) modified with a film of nickel tetrasulfonated phthalocyanine (p-NiTsPc) decorated with gold nanoparticles (AuNP). The modification process involved the potential cycling of the SPCE in NiTsPc and AuHCl4 solutions, leading to the rapid formation of spherical AuNP on the p-NiTsPc film surface. During the electrodeposition of p-NiTsPc, a characteristic redox couple at 0.43 and 0.53 V, attributed to NiII/NiIII from the O-Ni-O oxo-bridges, was observed. Subsequent electrodeposition of AuNPs resulted in a broad peak from -0.3 to -0.6V, which may be attributed to either the adsorption of gold precursor on the electrode surface or the nucleation of Au seeds. At more negative potentials (around -0.6 V), nucleation and particle growth occurred simultaneously. The p-NiTsPc film exhibited interesting behavior as a Raman/SERS probe and an electron mediator in voltammetric measurements of dopamine. Raman mapping performed on the SPCE/p-NiTsPc/AuNP platform showed homogeneous coverage of the SPCE surface with p-NiTsPc. The effective area of the SPCE/p-NiTsPc/AuNP electrode was higher than that of SPCE/p-NiTsPc and SPCE/AuNP, leading to increased charge transfer in the redox probe electrolyte [Fe(CN)6]3-/[Fe(CN)6]4-. The spectroelectrochemistry measurements of the SPCE/p-NiTsPc/AuNP platform, combining SERS and application of -60 mV, revealed a dopaquinone signal (oxidized species) and two characteristic vibrational modes of dopamine: 1270 and 1483 cm-1, ascribed to C-O stretching and catechol ring breathing, respectively. The results suggested that only the oxidized form of dopamine (dopamine-quinone) showed preferential adsorption at the film surface, allowing the Raman signal to be detected. Overall, the SPCE/p-NiTsPc/AuNP modified electrode exhibited potential applications as an electrochemical and SERS sensing assay, with a synergistic effect resulting from the role of p-NiTsPc in the electrochemical detection of dopamine and the AuNP in the enhancement of the Raman signal of the target molecule.
