Summary
Crystal violet (CV) is a synthetic basic cationic dye that stains violet (purple) in aqueous solutions. This molecule is applied as dye pigment in print inks and paints [1]. Detection of CV via surface-enhanced Raman scattering (SERS) is generally used to assess SERS substrate reproducibility due to its signal enhanced by the metallic nanoparticle. In this work, different aqueous concentrations of CV were utilized as a pollutant to be detected via SERS. The SERS technique can provide not only the fingerprint of the target molecule through its vibrational spectrum (selectivity), but also allows the detection of target molecules at low concentrations (sensitivity) [2]. The objective of this work was to detect the dye CV via SERS, comparing the performance of portable and benchtop Raman spectrometers and SERS liquid (colloid) and solid (paper-based) SERS substrates. CV aqueous solution on a concentration range from 1.0x10-3 to 1.0x10-7 mol L-1 were prepared. For SERS measurements using Ag colloid, the samples were prepared by mixing 990 µL of Ag colloid + 110 µL of CV, getting the final volume of 1.1 mL. For SERS measurements using paper-based substrate, the samples were prepared dipping the substrate in the analyte solution for 5 minutes, removing and letting it dry at room temperature. The laser line used was 785 nm for all the SERS measurements. The most intensity observed bands, at 420, 441, 525,723, 913, 1174, 1385, 1592 and 1620 cm-1 are attributed to vibrational modes of deformation and stretching CV ring, which suggest its adsorption on the surface of the Ag nanoparticles (AgNPs). The results of optimization show that both equipment allowed the detection of CV in the concentration range studied. Similar SERS spectrum profiles were obtained for both equipment regardless of concentration and both showed efficiency in detecting the SERS signal of the CV dye independent of the SERS substrates. Concerning the substrates comparison, solid (paper-based) SERS substrate showed the best CV SERS signal intensity for both equipment. No signal SERS were observed using Ag colloid for CV concentrations below 1.0x10-6 mol L-1 for both equipment. Reference: 1) de Voogt WP, editor.
