Resumo
1 INTRODUÇÃO Os metabólitos secundários produzidos por fungos filamentosos constituem importantes fontes de compostos bioativos com potencial para aplicação no desenvolvimento de medicamentos. Dentre esses compostos, destacam-se os antibióticos, que atuam por diferentes mecanismos de ação sobre os ácidos nucleicos, a síntese proteica, e na função da membrana plasmática. O primeiro medicamento considerado pioneiro no desenvolvimento de uma grande indústria para a descoberta e comercialização de novos antibióticos foi a penicilina, que demonstrou eficácia significativa no tratamento de doenças infecciosas. No entanto, o uso indiscriminado de antibióticos convencionais tem contribuído para o aumento da resistência antimicrobiana ao longo do tempo, sendo um dos principais desafios para a saúde pública mundial. Nesse sentido, os fungos do gênero Penicillium destacam-se como fontes promissoras de metabólitos bioativos. Dentre eles, Penicillium citrinum apresenta relevância biotecnológica devido à produção de compostos com atividade antimicrobiana pouco explorada frente a bactérias patogênicas de interesse clínico. 2 OBJETIVO Avaliar a atividade antibacteriana dos extratos extracelular e intracelular de P. citrinum frente às cepas Gram-positivas e Gram-negativas de interesse clínico, por meio da determinação da concentração inibitória mínima e concentração bactericida mínima. 3 METODOLOGIA Penicillium citrinum foi submetido à fermentação submersa em caldo Sabouraud por 28 dias para obtenção dos extratos extracelular e intracelular, os quais foram filtrados, concentrados e fracionados em acetato de etila. A atividade antibacteriana foi avaliada utilizando cepas da American Type Culture Collection: Escherichia coli, Staphylococcus aureus, Pseudomonas aeruginosa e Enterococcus faecalis. Os inóculos bacterianos foram padronizados para a concentração de 1,5 x 108 UFC/mL seguindo as recomendações do Clinical and Laboratory Standards Institute (CLSI, 2020). A atividade antibacteriana foi avaliada pelo método de microdiluição em caldo, utilizando placas de 96 poços, com determinação da concentração inibitória mínima (CIM) e da concentração bactericida mínima (CBM). Em cada poço, foram adicionados 100 µL dos extratos extracelular e intracelular de P. citrinum, preparados em caldo Mueller–Hinton contendo 0,1% de DMSO. As amostras foram testadas em concentrações variando de 1000 a 0,977 µg/mL, enquanto a gentamicina (32 a 0,5 µg/mL) foi utilizada como controle positivo. Após incubação por 24 h, foram adicionados 20 µL de resazurina a 1% e as placas foram reincubadas por 4 h. A leitura foi realizada visualmente, considerando-se a coloração azul como ausência de crescimento bacteriano e a coloração rosa como presença de crescimento. Para a determinação da CBM, alíquotas dos poços correspondentes à CIM e às concentrações superiores foram subcultivadas em placas de ágar padrão para contagem (PCA). Após incubação a 35 ± 2 °C por 24 h, a CBM foi definida como a menor concentração que apresentou = 20 UFC/mL. 4 RESULTADOS E DISCUSSÃO O extrato extracelular apresentou maior atividade inibitória frente a bactérias Gram-positivas, com valores de CIM de 10,40 µg/mL para S. aureus e 20,82 µg/mL para E. faecalis. No entanto, também foi observado efeito sobre a bactéria Gram-negativa E.coli (52,08 µg/mL). Em contraste, P. aeruginosa apresentou maior resistência, com CIM de 166,70 µg/mL, exigindo concentrações mais elevadas. O extrato intracelular apresentou atividade moderada em comparação ao extrato extracelular, com valores de CIM mais elevados, variando de 104,2 a 333,33 µg/mL para todas as cepas. Em relação à CBM, os valores para o extrato extracelular variaram de 15,625 µg/mL para S.aureus, 31,25 µg/mL para E. faecalis, 62,5 µg/mL para E.coli, e 250 µg/mL para P. aeruginosa. Já o extrato intracelular exibiu valores mais elevados, variando de 125 a 500 µg/mL. Para avaliação comparativa, a gentamicina foi utilizada. Como esperado, todas as cepas bacterianas mostraram-se sensíveis, com valores de CIM de 26,67 µg/mL para E. coli, 32 µg/mL para P. aeruginosa, 6,67 µg/mL para S.aureus e 13,33 µg/mL para E. faecalis. Em relação à CBM, as bactérias Gram-negativas (E. coli e P. aeruginosa) apresentaram valores semelhantes (32 µg/mL), enquanto as Gram-positivas exibiram valores distintos, com 8 µg/mL para S.aureus e 15,625 para E.faecalis. Os resultados evidenciaram maior atividade do extrato extracelular, principalmente contra bactérias Gram-positivas, com menor eficácia frente a P. aeruginosa, o que pode estar relacionado à maior complexidade estrutural de sua parede celular. Em comparação, a gentamicina apresentou maior atividade antimicrobiana em todas a cepas, confirmando sua eficácia. Ainda assim, os resultados reforçam o potencial de P. citrinum como fonte de bioativos antimicrobianos. 5 CONCLUSÃO O extrato extracelular de P. citrinum exibiu amplo espectro de ação antibacteriana, inibindo tanto bactérias Gram positivas quanto Gram negativas. Among these compounds, antibiotics stand out, which act by different mechanisms of action on nucleic acids, protein synthesis, and plasma membrane function. The first drug considered a pioneer in the development of a large industry for the discovery and commercialization of new antibiotics was penicillin, which demonstrated significant efficacy in the treatment of infectious diseases. However, the indiscriminate use of conventional antibiotics has contributed to the increase in antimicrobial resistance over time, being one of the main challenges for global public health. In this sense, fungi of the genus Penicillium stand out as promising sources of bioactive metabolites. Among them, Penicillium citrinum has biotechnological relevance due to the production of compounds with antimicrobial activity little explored against pathogenic bacteria of clinical interest. 2 OBJECTIVE To evaluate the antibacterial activity of extracellular and intracellular extracts of P. citrinum against Gram-positive and Gram-negative strains of clinical interest, by determining the minimum inhibitory concentration and minimum bactericidal concentration. 3 METHODOLOGY Penicillium citrinum was submitted to fermentation submerged in Sabouraud broth for 28 days to obtain extracellular and intracellular extracts, which were filtered, concentrated and fractionated in ethyl acetate. The antibacterial activity was evaluated using strains from the American Type Culture Collection: Escherichia coli, Staphylococcus aureus, Pseudomonas aeruginosa and Enterococcus faecalis. Bacterial inoculums were standardized to a concentration of 1.5 x 108 CFU/mL following the recommendations of the Clinical and Laboratory Standards Institute (CLSI, 2020). The antibacterial activity was evaluated by the broth microdilution method, using 96-well plates, with determination of the minimum inhibitory concentration (MIC) and the minimum bactericidal concentration (MBC). In each well, 100 µL of extracellular and intracellular extracts of P. citrinum were added, prepared in Mueller–Hinton broth containing 0.1% DMSO. The samples were tested at concentrations ranging from 1000 to 0.977 µg/mL, while gentamicin (32 to 0.5 µg/mL) was used as a positive control. After incubation for 24 h, 20 µL of 1% resazurin were added and the plates were reincubated for 4 h. The reading was performed visually, considering blue color as the absence of bacterial growth and pink color as the presence of growth. To determine the MBC, well aliquots corresponding to the MIC and higher concentrations were subcultured on standard agar plates for counting (PCA). After incubation at 35 ± 2 °C for 24 h, MBC was defined as the lowest concentration = 20 CFU/mL. 4 RESULTS AND DISCUSSION The extracellular extract showed higher inhibitory activity against Gram-positive bacteria, with MIC values of 10.40 µg/mL for S. aureus and 20.82 µg/mL for E. faecalis. However, an effect on the Gram-negative bacterium E.coli (52.08 µg/mL) was also observed. In contrast, P. aeruginosa showed greater resistance, with MIC of 166.70 µg/mL, requiring higher concentrations. The intracellular extract showed moderate activity compared to the extracellular extract, with higher MIC values, ranging from 104.2 to 333.33 µg/mL for all strains. Regarding MBC, the values for the extracellular extract ranged from 15.625 µg/mL for S. aureus, 31.25 µg/mL for E. faecalis, 62.5 µg/mL for E. coli, and 250 µg/mL for P. aeruginosa. The intracellular extract, on the other hand, showed higher values, ranging from 125 to 500 µg/mL. For comparative evaluation, gentamicin was used. As expected, all bacterial strains were sensitive, with MIC values of 26.67 µg/mL for E. coli, 32 µg/mL for P. aeruginosa, 6.67 µg/mL for S. aureus, and 13.33 µg/mL for E. faecalis. Regarding MBC, the Gram-negative bacteria (E. coli and P. aeruginosa) presented similar values (32 µg/mL), while the Gram-positive bacteria presented different values, with 8 µg/mL for S. aureus and 15.625 for E. faecalis. The results showed greater activity of the extracellular extract, especially against Gram-positive bacteria, with lower efficacy against P. aeruginosa, which may be related to the greater structural complexity of its cell wall. In comparison, gentamicin showed greater antimicrobial activity in all strains, confirming its efficacy. Still, the results reinforce the potential of P. citrinum as a source of antimicrobial bioactives. 5 CONCLUSION The extracellular extract of P. citrinum exhibited a broad spectrum of antibacterial action, inhibiting both Gram-positive and Gram-negative bacteria.
