Resumo
Some bacterial strains of the ESKAPE group (Enterococcus faecium, Staphylococcus aureus, Klebsiella pneumoniae, Acinetobacter baumannii, Pseudomonas aeruginosa and Enterobacter spp.) are drug multiresistant. Endolysins are proteins with potential for clinical use in treatments against resistant bacteria, due to their antimicrobial activity. For its application, one of the barriers to the exogenous use of endolysins against Gram-negative bacteria is the difficulty for crossing the outer membrane. SAR-endolysins are activated by pro-membrane destabilization in the outer membrane. Preliminary studies show that these proteins are able to penetrate the outer membrane. In addition, several compounds have the ability to destabilize both cytoplasmic and outer bacterial membranes, and already shown synergy with endolysins against Gram-positive and Gram-negative bacteria. Thus, the objective of this project is the recombinant expression and purification of a phage LysM SAR-endolysin, in silico analysis, and investigating their antibacterial activity, alone or combined permeabilizing agents, against bacteria of ESKAPE group through in vitro and in vivo assays in the Galleria mellonella model. We are performing expression and protein purification, however we have already analyzed the LysM SAR-endolysin by ProtParam tool which identified a molecular weight of 47263.73 Daltons, has a theoretical half-life higher than 10 hours inside cells like Escherichia coli, instability index of 15.15 classifying this protein as stable; GRAVY index of -0.323 indicating that the protein is hydrophilic. The theoretical isoelectric point is 6.03, calculated using the Isoelectric Point Calculator 2.0 (IPC 2.0) web server. Therefore, we concluded that LysM is stable and hydrophilic.
