Resumo
The Brazilian campos rupestres, a biodiversity hotspot, is an ecoregion that frequently experiences prolonged droughts. Plants that inhabit this environment have developed various strategies to deal with water scarcity, particularly those from the Velloziacea family. In order to gain an understanding of the mechanisms involved in these diverse drought-coping strategies, we selected two representative species from the Vellozia genus: the desiccation-tolerant V. nivea and the evergreen V. intermedia. To study the molecular basis of these phenotypes, we focused on the OSCA gene family, which encodes transmembrane proteins known as OSCA channels. These channels play a role in increasing [Ca2+]i levels in response to osmotic stress in plants. Differences in gene expression patterns related to OSCA may indicate that the two species employ distinct drought perception mechanisms. To further investigate this, we collected 80 samples comprising leaf and root tissues from both species and exposed them to four different water regimes: early drought (ED), late drought (LD), early rain (ER), and late rain (LR). We then used weighted gene co-expression network analysis (WGCNA) to generate gene interaction networks from the dataset. By characterizing the modules of interest based on their expression patterns, and identifying the OSCA-connected genes within these modules, we were able to identify functional enrichment of these genes. We found 13 modules (8 for leaf and 5 for root) with an average of 300 genes per module and 4 modules (3 for leaf and 1 for root) with an average of 3200 genes per module that are associated with OSCA expression in V. intermedia and V. nivea, respectively. Furthermore, some of these modules were enriched in functions related to abiotic stress response, such as nitric oxide and reactive oxygen species responsiveness, as well as photosynthetic metabolism in the leaf. Additionally, we observed that the evergreen V. intermedia had a higher degree of modularity compared to V. nivea, indicating a more complex transcriptional regulatory network is required for it to survive under varying water availability. By utilizing cutting-edge gene network modeling, we were able to identify key interactors associated with OSCA genes in the different strategies for coping with drought. Our findings on the transcriptomic landscape of Vellozia spp. genes may provide new insights into how wild plants respond to drought, which is a pressing concern in a world facing the challenges of climate change.
