Resumo
To produce ethanol, Clostridium thermocellum, a thermophilic, anaerobic bacteria, uses a complex and multiplex metabolic pathway that uses the pyruvate ferredoxin oxidoreductase (PFOR) enzyme. However, the ethanol titer produced by this organism is too low for commercial applications. Most commercial ethanol production relies on a much simpler metabolic pathway that uses the pyruvate decarboxylase (PDC) enzyme, which is found in both bacteria and yeast (e.g. Saccharomyces cerevisiae), but does not function at thermophilic temperatures. Previous attempts to introduce PDC activity into thermophilic bacteria, such as C. thermocellum, have not been successful. The aimed pathway involves PDC and alcohol dehydrogenase (ADH) to convert pyruvate to acetaldehyde and then ethanol. This pathway also converts NADH to NAD, so for in-vitro reactions, we often include a third enzyme, formate dehydrogenase (FDH) to recycle the resulting NAD back to NADH. Aiming to build a computational model that reflects the actual pathway in E. coli and help troubleshoot the C. thermocellum cell free system to produce high ethanol titer, my goal is to demonstrate that the in-vitro PDC-ADH-FDH system is capable of producing high concentrations of ethanol (> 1 M) and determine the extent to which this behavior can be mathematically modeled using existing kinetic models.
