Resumo
Salmonella enterica is a gram-negative bacterium that can infect humans and animals used in the food industry. S. enterica has an important role in the problem of emerging antibiotic resistance. Bad administration of antibiotics has pushed the use of last-resort antibiotics, such as colistin. Yet, the continued use of colistin in animal husbandry has led to the emergence of mobilized colistin resistance genes (MCRs). In light of that, curated databases enable the study of the evolutionary events and genomic context of elusive genes such as the mcr, which are mostly found on plasmids. Here, we provide a pangenome analysis of 1544 Salmonella enterica genomes from a curated database, including mcr-positive and mcr-negative samples. We found a prevalence of mcr-9, followed by mcr-1 among the samples. Further analysis of genomic distances revealed a tendency for accessory distances among mcr-positive samples to be smaller than between mcr-positive and negative samples, suggesting a more similar composition. Moreover, we analyze the pattern of association and dissociation between genes in the pangenome, and after inferring the location of the genes that associate and dissociate with mcr genes, a pattern of association linked to plasmids and dissociation linked to chromosomes emerged. Further, functional evaluation of mcr-associated and dissociated genes revealed that viral proteins could take part in both association and dissociation processes, which could be pointing to different mobile elements carrying mcr genes. Also, we screened the IGRs related to mcr genes and found already-known IS sequences related to them. These findings point to a dynamic in the genetic context of mcr genes influenced by horizontal transmission. Moreover, it sheds light on the antagonistic relationship between the two more common mcr genes on the dataset, mcr-1 and mcr-9 on Salmonella enterica. Fomento: CAPES
