Resumo

Salmonella enterica and Shigella flexneri are part of the major foodborne pathogens associated to diarrheal diseases and systemic infections. The treatment of these infections is usually based in antibiotics that together with its misuse has led to the rise of antibiotic resistant and multi-resistant bacterial strains. That’s the reason why there is an increasing need to finding novel drug therapies and a better understanding of pathogenesis mechanisms and regulatory proteins can be a helpful tool in the comprehension of antibiotic resistance mechanisms as well as the discovery of new therapies targets. That’s why the aim of this project is to study the function of regulatory proteins in Salmonella and Shigella trough the genic deletion and molecular and phenotypic analysis. The attenuation of mutant strains will be assessed using the C. elegans and G. mellonella alternative animal models. Furthermore, we will try to clarify the role of HU protein in the physiology of Shigella flexneri as well as proposed it as a potential target for the development of new therapies. Finally, will be made complementary studies of ProQ protein in Salmonella enterica. Preliminary results shown that ?proQ mutants are attenuated in the C. elegans and G. mellonella models.
