Summary
Micronutrients present antioxidant activities related to the improvement of immunity and possible antitumor effects. Since cancer is a multifactorial disease, these substances are studied to prevent or treat it, presenting discordant data in the literature. In this sense, the immunomodulatory properties of a-tocopherol and selenomethionine were studied in Ehrlich tumor model, by evaluating the cytokines and nitric oxide (NO) production of splenocytes at the basal condition or under tumor influence. Spleen cells from healthy animals (1x106 cells/mL) were maintained in culture with medium without and with ascites (30% off the well volume) for 48 hours. The culture was supplemented with the micronutrients, isolated or combined at the concentrations of 15, 31, 62, 125, 250 and 500 µg/mL. Lipopolysaccharide (10 µg/mL) was used as activation control. NO was determined by the Griess method and cytokines by flow cytometry. Supplemented splenocytes increased the production of IL2, IL4, IFN-gamma, TNF- alpha, IL6 and IL10 when compared to the basal condition. Isolated micronutrients induced an elevation in IL2 production greater than LPS stimulated cells (5.62 vs 3.8 pg/ml). When cells were cultured with ascites fluid, IL4, IFN-gamma, TNF-alpha and IL6 increased. However, the presence of micronutrients in this context caused the decrease of IL6. In the presence of ascites, splenocytes produce more NO than cells maintained only in medium, but, in the presence of the associated micronutrients (500 µg/mL), there was a return to the basal concentration. Our data allow us to conclude that these micronutrients are capable of modifying the production of cytokines, especially IL2, important for its lymphoproliferative function, suggesting a favorable action on the immune response. In the presence of the tumor, the micronutrients reduce inflammatory mediators such as IL6 and NO, suggesting that the antitumor effect can be through the modulation of the inflammatory response.
